If it's for The Tharn Speaks it's either in Grahtwood, Deshaan or Stormhaven depending on your alliance (resp. AD, EP or DC).

The map marker? The marker pins the location where you gotta go, I've never had a marker showing up at a wrong location in all my game time.

What happens with us lesser mortals is that we don't plan our ultimate route ourselves but look at the map and head towards the white arch thingy.
This tells us that you need to use that passage way to help you get to where you want to go (whether its a boat or caravan or just a gate in the road). Often, though, those markers are either plain wrong or take you round in circles. I learned this the hard way and always plan my own route now.

As I said, I go to a marker for a quest. And many time it tells me I need such and such a DLC to go there. One such is Daggerfall, and I don't see that DLC anywhere. If I go to the world map, I can see the other regions but I can't fast travel there because I haven't discovered any Wayshrines there(not having been there yet). So, do I have to physically travel there? Can I get to those regions on foot?
Yes, if you don't have the wayshrines you either go there on foot or boat or caravan. As for Daggerfall it's near the harborage for DC players, in Glenumbra, the first big city you'll see.
Edit: You'll go there eventually though, once you finish quests in the Glenumbra zone, you'll be sent to Stormhaven, then Rivenspire, then the Alik'r Desert and finally Bangkorai, if I'm not mistaken.
Since you're AD though, you're looking for a place in Grahtwood most likely, not Daggerfall or anything in that part of the map.

I've found that, sometimes, there are ways to get to parts of the main story quests that are also part of a DLC, but there are other ways to get to that location that don't require it. ESO could improve their guidance in that respect.

I quite enjoy travelling to these places on foot/mount. If you aren't in a hurry you pick up some adventures along the way. Use the world map so you know what general direction you need to head off in. You can usually tell the road you need because it goes furthest towards where you're going.
Use Mighty Jean's directions then go a a wayshrine. If you look at all the Glenumbra map the quest marker should be there for you. Port to the nearest wayshrine - even if it's in a different zone and then trudge the rest of the way, using the world map for general direction guidance.
According to the other thread about the main quest OP is a wood elf, so from AD. Harborage should be in Auridon and the delve for The Tharn Speaks in Grahtwood (near the north-western exit, can't recall the english name... Something-something depths ?).
I think there's no other quest from the Prophet that leads you that far from the Harborage, they're either an instance accessed from the Harborage or near the very first town in the first zone. But it's been a long time so I may not remember it correctly.
Yea, I don' t recall having this problem back when we don't have these many DLC and I seen a lot people complain about this. Now that I made another toon I finally understand.
The marker will take you to the DLC if you are not in the zone of the quest, the DLC area however has a boat/carriage that will take you to this zone. Stupid bug that has not been fixed for long time.
Just follow what they said above, or look for the quest guide in google, I know it's a pain, but well..
I found Auridon. It's way in the SW of the map and I didn't see it at first glance since most of my questing was done in the NE part of the region map. I'm there now and working on quests which i'm sure will lead to other areas. For example, there is a boat that will take me to Ghratwood when I'm ready.
